sister chromatid exchange analysis in some holstein bulls

چکیده
chromosomal appearance of 12 holstein bulls selected for artificial inseminations were examined bysister chromatid exchanges (sces). for differential staining of sister chromatids bromodeoxyuridine (brdu)was inoculated in lymphocyte cultures. the mean, maximum and minimum number of sce per cell weredetermined 6.8 ± 1.14, 8.3 ± 1.1 and 5.7 ± 1.5, respectively. sce frequencies of all animals were in thenormal level. also, evaluation of different breeding condition and age range did not show any significantstatistical (p>0.05) effect on sces rates

Sister chromatid exchange (SCE) techniques have been shown to have great potential for use in the in vitro screening of suspected mutagens and carcinogens. Application of these techniques to bone marrow cells and/or lymphocytes from patients with various hematologic malignancies has demonstrated significant abnormalities in SCE frequencies in some of these diseases, as well as showing drug trea...

Sister chromatid exchange (SCE) frequency in chromosomes from lymphocytes of a patient with dyskeratosis congenita was 12-2 per mitosis. Our 33 normal controls had a mean of 5-4 SCE per mitosis and 5 patients with Fanconi's anaemia averaged 7-6 SCE per mitosis. The rate of chromosome breakage was only 0-5% in the dyskeratosis congenita patient and 0 to 2-5% in controls, while the Fanconi's anae...

Sister-chromatid exchanges (SCE) are the result of apparent exchanges of DNA between chromatids and may be visualized in metaphase chromosomes after growth of cells for 2 cycles in bromodeoxyuridine and appropriate staining procedures. The frequency of SCEs is thought to be a sensitive indicator of genetic instability and early chromosome changes (Perry and Evans, 1975; Solomon and Bobrow, 1975...
متن کاملAutomatic measurement of sister chromatid exchange frequency.
An automatic system for detecting and counting sister chromatid exchanges in human chromosomes has been developed. Metaphase chromosomes from lymphocytes which had incorporated 5-bromodeoxyuridine for two replication cycles were treated with the dye 33258 Hoechst and photodegraded so that the sister chromatids exhibited differential Giemsa staining. A computer-controlled television-microscope s...
متن کاملThe sister chromatid exchange (SCE) assay.
A fully optimized staining method for detecting sister chromatid exchanges in cultured cells is presented. The method gives reproducibly robust quantitative results. Sister chromatid exchange is a classic toxicology assay for genotoxicity and for detecting alterations to the biochemistry underlying cellular homologous recombination. Growth of cells in the presence of 5'-bromo-deoxyuridine for t...
